The main physiologic defect in XLH is impaired proximal renal tubular reabsorption of phosphate, which is crucial to healthy growth and development, bone formation, acid-base regulation, and cellular metabolism. Skeletal manifestations in adults and children are debilitating and significantly impact quality of life. Breakthrough therapies have emerged to improve the efficacy and tolerability that current treatments severely lack.
